Neurogenic influences of bilateral adrenalectomy on monoamine oxidase

Summary

Bilateral adrenalectomy increased monoamine oxidase activity in several rat organs, primarily due to neuronal influences. This suggests neuronal origin, not transsynaptic induction, for the enhanced enzyme activity.

Background:

Bilateral adrenalectomy in rats leads to increased monoamine oxidase (MAO) activity in specific organs.
The precise origin and localization (intra- or extraneuronal) of this enhanced MAO activity remain unclear.

Purpose of the Study:

To investigate the neurogenic influences responsible for the increased MAO activity post-adrenalectomy.
To determine whether the enhanced MAO activity is located within or outside neurons.

Main Methods:

Bilateral adrenalectomy in rats.
Administration of ganglionic blocker chlorisondamine.
Surgical denervation of the superior cervical ganglion.
Treatment with 6-hydroxydopamine to assess neurogenic contribution.

Main Results:

Adrenalectomy increased MAO activity in the heart, vas deferens, spleen, superior cervical ganglion, and hypothalamus.
Ganglionic blockade and surgical denervation did not affect the rise in MAO activity.
6-Hydroxydopamine abolished the increase in MAO in the vas deferens, spleen, and superior cervical ganglion, indicating a neuronal origin.

Conclusions:

The increase in organ monoamine oxidase activity following bilateral adrenalectomy is predominantly of neuronal origin.
The observed enhancement of MAO activity is not a result of transsynaptic induction.
